  Description: 

Oligo(dT)12-18 Primer hybridizes to the poly(A) tail of mRNA and is suitable 

for use as a primer for first stand cDNA synthesis with reverse transcriptase. 

The primer is supplied in DEPC-treated water at a concentration of 0.5 μg/μl. 

The primer is phosphorylated at the 5' end. 

Component: Oligo(dT)12-18 Primer 

Recommended Amount: We recommend using 0.5 μg of oligo(dT)12-18 primer per μg of poly(A)+

RNA. For total RNA, we recommend 0.5 μg of primer for < 5 μg of RNA. 

Functional Assay Conditions :

1 μl oligo(dT)12-18 primer (0.5 μg/μl) 

1.0 μg poly(A)+ RNA 

Sterile distilled water 

Heat mixture to 70°C for 10 min, and quick chill on ice. 

4 μl 5X first strand buffer [250 mM Tris-HCl (pH 8.3), 375 mM KCl, 15 mMMgCl2] 

2 μl 0.1 M DTT 

1 μl dNTPs (10 mM each dNTP) 

1 μl [α-32P]dCTP (1 μCi/μl) 

200 units SuperScript? RT (Cat. No. 18053-017) or M-MLV RT (Cat. No.28025-013/28025-021) 

Total volume: 20 μl 

Incubate at 37°C for 1 h. 

Quality Control :This product is qualified in a first strand cDNA reaction.
